IGL Tube – 6FAM for ABI Fluorescence Detection is a Research Use Only master mix that can be used to identify immunoglobulin lambda (IGL) light chain gene rearrangements.

    The human immunoglobulin lambda (IGL) light chain locus is located on the long arm of chromosome 22 (22q11.2) and spans 1050 kilobases.  It is made up of 73-74 variable (Vλ) gene segments (spread over 900 kilobases), 7-11 joining (Jλ) gene segments and 7-11 constant (Cλ) gene segments depending on the haplotypes. Of the 73-74 Vλ region genes, only 30-33 are functional and can be grouped into 11 families and 3 clans.  The Jλ and Cλ region genes are organized in tandem with a Jλ segment preceding a Cλ gene.  Typically there are seven (7) Jλ-Cλ segments of which four (4) are functional and encode the four (4) Ig lambda isotypes. Vλ-Jλ rearrangements potentially represent an attractive extra PCR target for clonality studies to compensate for false-negative IGH VH-JH PCR results mainly caused by somatic hypermutations.

    The IGL Tube master mix targets conserved regions within the variable (Vλ1-3) and the joining (Jλ1-3) regions that flank the unique, hypervariable, antigen-binding, complementarity determining region 3 (CDR3).  Clonality is indicated if the master mix generates clonal band(s).  PCR products are differentially labeled with 6FAM and can be analyzed using any platform that has the ability to detect these fluorochromes.
