IGK Tube B – 6FAM for ABI Fluorescence Detection is a Research Use Only master mix that can be used to identify immunoglobulin kappa (IGK) light chain gene rearrangements.

    The human immunoglobulin kappa (IGK) light chain locus on the short arm of chromosome 2 (2p11.2) spans 1820 kb.  It is made up of 76 variable (Vκ) gene segments belonging to 7 subgroups, 5 joining (Jκ) gene segments, and one constant (Cκ) gene segment.  Productive assembly of the kappa gene is successful in about 60% of human B lymphocytes.  However, even when unsuccessful, clonal B-cells generally retain the rearranged kappa genes.  The Vκ segments encode the first 95 N-terminal amino acids.  Positions 96-108 are encoded by one of five joining (Jκ) gene segments.  The constant (Cκ) portion of the kappa light chain (amino acids 109-214) is encoded by a single constant (Cκ) region separated from the Jκ region by an intron.  The length of the hypervariable complementarity determining region 3 (CDR3) in kappa light chain genes is limited and rearrangements in this region display significant skewing (platykurtosis).

    This master mix targets conserved regions within the variable (Vκ1-7) and the joining (Jκ1-5) regions that flank the unique hypervariable, antigen-binding, complementarity determining region 3 (CDR3) as well as the Kde and intragenic regions.  IGK Tube B contains six upstream Vκ region primers, an upstream intragenic primer and a downstream Kde primer.  Clonality is indicated if the master mix generates clonal band(s).  PCR products are differentially labeled with 6FAM and can be analyzed using any platform that has the ability to detect these fluorochromes.
