LeukoStrat® KMT2A + MRD Assay

Assay Use

The LeukoStrat® KMT2A + MRD Assay targets the KMT2A gene to identify four AML-related KMT2A rearrangements (KMT2Ar): MLLT3 (AF9), MLLT10 (AF10), ELL, and MLLT4/AFDN (AF6).

  • Specimen Requirements

    This assay tests ribonucleic acid (RNA) isolated from peripheral blood or bone marrow. Assay sensitivity is dependent on RNA input. 

    Screening requires a minimum input quantity of  100 ng of high quality RNA for a sensitivity of 5×10-3 (0.5%).

    Measurable residual disease testing input is dependent on the desired sensitivity. The assay’s maximum input of 5 µg of high quality RNA achieves a sensitivity of 5×10-5 (0.005%).

  • Description

    The  LeukoStrat® KMT2A + MRD Assay is a digital PCR-based assay that simultaneously detects four AML-related KMT2A gene rearrangements (KMT2Ar) – MLLT3 (AF9), MLLT10 (AF10), ELL, and MLLT4/AFDN (AF6). 

    The comprehensive kit includes a primer probe mix and all necessary controls, and has a dynamic RNA input range for both  screening and  measurable residual disease detection at a sensitivity down to 5×10-5.

    Designed for use with the Roche Digital LightCycler® System, the  companion LeukoStrat® KMT2A + MRD Software uses the system’s raw data files to determine and report the presence of KMT2Ar and the KMT2Ar:ABL1 ratio. This normalization against a stable control gene enables reliable, accurate quantitation, supporting confident decision making.
