FLT3 D835 Master Mix - 6FAM for ABI Fluorescence Detection
FLT3 D835 – 6FAM for ABI Fluorescence Detection is a Research Use Only master mix that can be used to identify FLT3 D835 mutations.
FLT3 is a receptor tyrosine kinase that is normally expressed on many cell types including hematologic stem cells. Mutation of the FLT3 receptor, by either internal tandem duplication (ITD) of the juxtamembrane domain or point mutation in the activation loop of the tyrosine kinase domain (TKD), causes constitutive activation of the FLT3 receptor. The most prevalent type of FLT3 mutation is an internal tandem duplication in and around the juxtamembrane domain. The second most common mutation type in the FLT3 gene is a TKD point mutation in aspartate (D835) or isoleucine (I836).
Detection of the D835 mutation requires digest with EcoRV enzyme (not included). After digestion, the wild-type FLT3 produces three (3) bands of 21, 49, and 80 bp. The D835 mutant loses one restriction site and generates a 129 bp band and a 21 bp band. The presence of a FLT3 D835 mutation is indicated if, after EcoRV digestion, a129 bp band is generated. PCR products are differentially labeled with 6FAM and can be analyzed using any platform that has the ability to detect these fluorochromes.
